An agreement could be made between 2 teams beforehand though. "If Mariota is on the board we'll trade this..." kind of thing.

Totally agree the teams would make those agreements but their execution would only occur when the pick is "on the clock".

I still have serious doubts that the Titans are not going to take Marcus Mariota. If they don't then I think they will take the picks to move back. The Titans are the one team that could take Marcus Mariota and then perhaps put him up for auction afterwards.

They wouldn't.  But the point is that they cannot sit back and make plans to trade to #4 or #5 without taking the chance that someone who wants him moves up to #3.  If they really want to secure it, they need to not leave those gaps.
