Yes or at least have that built into your offensive scheme beforehand. This is hypothetical and an extremely simplified case, but imagine your running through your C gap on the right side with amazing success in the first half. To not expect the opposing teams defense to adjust, whether that is defensive line adjustment or a LBer lining up for that particular gap, you have to have an alternate playcall. Continuing to run through that C gap with no change will certainly result in less success.
